- Shows tranylcypromine irreversibly inactivates LSD1 by covalently modifying its FAD cofactor, while inhibiting MAO-B and MAO-A more potently than LSD1.
- Identifies tranylcypromine as the most potent LSD1 inhibitor among the MAO inhibitors tested. In P19 embryonal carcinoma cells, it raised global H3K4 methylation and switched on LSD1 target genes, including Oct4.
- Combined with the GSK-3 inhibitor CHIR-99021, tranylcypromine (Parnate) enabled reprogramming of human primary keratinocytes with only Oct4 and Klf4, the first human iPSCs generated from somatic cells without exogenous Sox2.
