- Identifies RG108 as a DNMT inhibitor that blocks enzyme activity without trapping the enzyme on DNA, demethylating genomic DNA at low micromolar concentrations without detectable toxicity and reactivating silenced tumor suppressor genes.
- Demonstrates that RG108 treatment reprogrammed mouse skeletal myoblasts into iPS-like cells without exogenous transcription factor delivery, with resulting cells expressing pluripotency markers and exhibiting cardiomyogenic differentiation potential.
- In human bone marrow mesenchymal stem cells, 50 µM RG108 reduced DNMT activity and global DNA methylation without affecting viability, and epigenetically activated NANOG and OCT4 at their promoters.
